PCR LIGHTCYCLER CHECK SHEET

MEDICAL SYSTEM LIGHTCYCLER (PCR) SYSTEM

1. Confirmatory Laboratory
2. Is there at least one 8506 with PCR-certification from BDRD onboard?
3. Does the PCR-trained technician have a SOP binder containing Sections 1-8 and Section 11?
4. Analyzer and Laptop:
a. Does the laptop (or alternate computer) boot up without problem and contain applicable LightCycler 3.3 or RAPID 1.2.14 software?
b. The laptop’s hard drive should be no more than 70% full. (If the hard drive is partitioned, check the drive that the analyzer’s software is running off of.) The laptop should contain no personal information or files that do not directly pertain to the Confirmatory lab.
c. With the laptop attached, does the analyzer turn on and perform a self-test (if LightCycler) or warmup run (if RAPID)?
5. Freezer:
a. Are the reagents stored in a -20°C manual defrost freezer?
b. Is a daily temperature log maintained on the freezer, with the average temperature over the last 3 weeks being no higher than -10°C?
6. Bio-Safety Hood:
a. Is there a bio-safety hood installed on the ship, with certification performed within past 12 months?
7. Consumable Supplies:
a. Between the “Clean” and “Dirty” areas, does the Confirmatory lab have a minimum of the following critical supplies:

a. Does the lab have a total of five colored boxes** (3 boxes of Master Mix, 2 boxes of Standards) and one box of QC samples, stored in the appropriate freezer and containing a minimum of the following:

c. 9 tubes of each target Master Mix (14 target assays in kit)
d. 12 tubes of Inhibition Master Mix

g. Standards and Positive Controls: 2 boxes
h. 1 tube of 100,000 C Positive Control stock for each assay (14 target assays in kit)
i. 3 tubes of Spike Positive Control
