Enhancer predication in 8q24 cancer-associated regions. Enhancers were predicted with EEL. Sequences for 8q24 regions and their dog and mouse orthologs were downloaded from UCSC genome website ( Region1 and region3 were predicted by EEL together. Region2 was predicted separately from region1 and region3. The top candidates in both human-dog alignments and human-mouse alignment were compared to each other. The top 4 candidates in region1 and region3, and top 3 candidates in region2 were picked for further analysis. Their positions and ranks are listed in supplementary table. Supplemental Figure. 1. Effect of CMV Enhancer Element over Myc and TK Promoters. The CMV enhancer was cloned in front of a Myc promoter-driven or TK promoter-driven luciferase-GFP reporter. Luciferase activity was recorded in prostate cell lines LnCAP, and PC3 as well as colorectal cell line SW620. Enhancer reporter plasmids were co-transfected into cell lines as described in materials and methods. Luciferase activity was measured 24 hrs post-transfection using the Dual-Glo Luciferae assay system.
