sTarting information
You need information about:
· Column
· Flow Rate
· UV detection
· Solvent and Gradient
1) Start Up system
Click on “SPX” in the touch screen for start the software
Click on “Chemistry”
2) Method editor
If you use a method:
Select a “User”
Select a “Method”
Select “Sample name” : give a name
If you need a new method Select “Wizard” or “New”
- Separation :
Weak solvent: Select a solvent (if you don’t find your solvent click “Show all”) A
Strong solvent: Select a solvent B
Cartridge Type: Select (See manual page 5)
Flow rate: Select (See manual page 5)
- UV detector
Select collection wavelength (nm)
Select monitor wavelength (nm)
- Collection Mode
Select a collection mode (See manual page 7)
- Rack allocation
Rack type: select a rack (See manual page 9)
Dispense order
Max flow volume (check your tubes!)
- Edit gradient
(The gradient table is based on solvent B)
If you wish “initial waste” click and enter the value for Initial waste (ml)
If you wish to equilibrate click “Equilibration time”:
Start: the percentage of strong solvent B at the start of the equilibration
End : the percentage of strong solvent B at the end of the equilibration
Length : enter the equilibration volume (CV column volume or ml)
Click “Add After”:
Start: the percentage of strong solvent B at the start of gradient
End : the percentage of strong solvent B at the end of gradient
Length : enter the volume (CV column volume or ml)
If you wish add more step
3) Run a purification
Click “Status”: click “lamp ON” (wait for 10 min)
Connect the bottles of solvent to the system
Click “Solvent”:
Select “Prime”
Select “Trough column”
Select first solvent concentration 100%
Select a volume (es. 60 ml)
Select flow rate (es. 10 ml/min)
Click “Prime”
Repeat the operation for the second solvent
Click “Finish”
Click the GREEN ARROW in the method Editor
Install the column on the system
Select “Equilibration Time” : wait the finish of equilibration
Select “Load sample” : load the sample in the column
Select “Run Gradient” : the purification starts
