Annexe 1: List and composition of the media used
The following media (quantities are given in g.l-1 unless otherwise stated, and solidified with 15 g.l-1 agar when necessary) were used.
- BG-11. Stock solution # 1 {Na-EDTA, 0.1; Ferric ammonium citrate, 0.6; citric acid.1H2O, 0.6; CaCl2.2H2O, 3.6}. Stock # 2 {MgSO4.7H2O, 7.5}. Stock # 3 {K2HPO4.3H2O, 4}. Stock # 4 {NaNO3, 150}. Stock # 5 {B & D micro-nutrients – see Broughton and John (1979)}. Basic BG11 comprised 10 ml.l-1 of Stocks # 1 to 4, 1 ml B & D micro-nutrients and 0.02 g Na2CO3 (pH 7.5). Stock # 3 was omitted in N-free BG11.
- Czapek-Dox (CzD){sucrose 30; NaNO3 3.0; MgSO4.7H20, 0.5; KCl 0.5; FeSO4.7H2O, 0.01, K2HPO4, 1 ( pH 7.3)}.
- DG18 {peptone 5, KH2PO4, 1, 2,6-dichloro-4-nitroanilide 0.002, glycerol 200, glucose 10, MgSO4, 0,5 g chloramphenicol 0,1 g (pH 5.6)}.
- DRBC {proteose peptone, glucose, 10; KH2PO4, 1; MgSO4, 0.5 g; dichlorane, 0.002, Rose Bengal, 0.025, chloramphenicol, 0.1 (pH 5.6)}
- Erythritol-chloramphenicol agar (ECA){meso-erythritol, 10; chloramphenicol, 0.02; yeast-nitrogen base (Difco), 11.7 (pH7.0)}.
- Maxted {proteose peptone, 5; yeast extract, 3; 0.2 Na2HPO4, 2; glucose, 2)}.
- Malt-extract agar (MEA){malt extract, 20; peptone, 1; glucose, 20)}.
- Modified Bold’s Basal Medium (MBB –Friedl, 1989){Macro-nutrients (NaCl, 2.5; CaCl2.2H20, 2.5; KNO2, 50: MgSO4.7H2O, 6.5; (NH4)HPO4, 25). Potassium (KOH, 30; EDTA, 50). Iron (FeSO4.7H2O, 5). Borate (H3BO3, 11.04). Micro-nutrients (ZnSO4.7H2O, 8.82; MnCl2.4H2O, 1.44; NaMoO4, 0.72; CuSO4.5H2O, 0.158; Co(NO3).6H2O, 0.5). Vitamins (thiamine, 10; biotin, 0.1; vitamin B12, 0.02; nicotinamide, 0.01). Basic MBB comprised 10 ml.l-1 of each solution (except micro-nutrients, 1 ml), and 0.715 g HEPES (pH 5.5). For heterotrophic algae 15 g glucose and 20 g proteose peptone were added to the above (pH 7.1).
- Plate-count agar (PCA){casein-peptone, 5; yeast-extract, 2.5; glucose, 1 (pH 7.0)}.
- PYGV {peptone, 0.2; yeast-extract, 0.2; glucose, 0.2; Hunter’s mineral solution (nitriloacetic acid, 10; MgSO4.7H2O, 29.7; CaCl2.2H2O, 3.3; NaMoO4.2H2O, 12.7; FeSO4.7H2O, 0.4; EDTA, 0.13; ZnSO4.H2O, 0.88; CuSO4.5H2O, 0.02; CoCl2.2H2O, 0.01; Na2B4O7.10H2O, 0.009), 20 ml; vitamin solution (biotin, 0.004, pyridoxine-HCl, 0.02; thiamine-HCl, 0.02; Ca-pantothenate, 0.01; folic acid, 0.004; riboflavin, 0.01; nicotinic acid, 0.01, vitamin B12, 0.0002), 10 ml (pH 7.0)}.
- R2A {proteose peptone, 0.5; casamino acids, 0.5;
yeast extract, 0.5; glucose, 0.5; soluble starch, 0.5; K2HPO4, 0.3; MgSO4.7H2O, 0.05; Na-pyruvate, 0.3 (pH 7.2)}. - Tryptone Soya Agar (TSA){casein peptone, 15; soya peptone, 5; NaCl, 5; (pH 7.3)}.
- YM3 Agar {CaCO3, 0.1; K2HPO4, 0.3; MgSO4.7H2O, 0.1; MnSO4, 0.01; NaCl, 0.1, NH4Cl, 0.05; urea, 0.03, mannitol, 5.0; sucrose, 5.0; yeast extract 2.5 (pH 6.8)(Broughton and John, 1979)}.
