Intro to DNA Fingerprinting

BACKGROUND: A health inspector went to a McDonald’s and found a piece of hair on an Egg McMuffin! The manager needs to find out who he will fire so he sent the hair and his workers’ hairs to Westlake High School for testing. Luckily, Mrs. State was able to extract the DNA from the pieces of hair as well as from the 5 McDonald’s breakfast crew members.

Here are the DNA samples that Mrs. State extracted: 1.Source DNA (from the hair on the Egg McMuffin)

  1. Crew Member #1 – Annie
  2. Crew Member #2 – Becky
  3. Crew Member #3 – Charles
  4. Crew Member #4 – Darrel
  5. Crew Member #5 – Elliot

Now that Mrs. State has extracted the DNA, you have been asked to figure out which of the crew members was responsible for the hair found on the Egg McMuffin. The only “tools” that you have are threedifferent restriction enzymes(RE) that recognize and cutthree different DNA sequences. You also have another tool that separates different pieces of DNA by how long the DNA strands are. (Note: you do not have the technology to actually “read” the DNA sequences.)

**These restriction enzymes make blunt cuts, not jagged… so no sticky ends…how fortunate!!**

Purpose: To become familiar with the process of DNA fingerprinting.

Directions: Follow the procedures on the back to create a poster that shows how a DNA fingerprint is made.

Intro to DNA Fingerprinting

PROCEDURES:

Read ALL the directions and follow them step-by-step to create a poster that displays your evidence.

  1. Write the title (Example: There’s a Hair in my Egg McMuffin!) at the top of your poster.
  2. Write all the names of you and your group members on your poster.
  3. On each DNA strand, look for these sequences, then draw a box around them, where the restriction enzymes will cut:

  1. Prepare yourevidence.
  2. For the Source DNA, cut in the middle of the boxes you drew (the dotted lines above)… don’t lose any pieces!!!
  3. Arrange the Source DNA strand by length, in a row. Longest pieces should be on the left. Leave about an inch of space between each piece.
  4. Glue the Source DNA pieces onto your poster, just as you arranged it. Label it “Source DNA.” Leave room to glue the DNA strands from the other crew members below.
  5. ForAnnie’s (Crew Member #1) DNA, cut in the middle of the boxes you drew (the dotted lines above)… Don’t lose any pieces!!!
  6. Arrange Annie’s DNA strand by length, in a row. Longest pieces should be on the left. Leave about an inch of space between each piece.
  7. GlueAnnie’s DNA pieces onto your poster, just as you arranged it. Label it “Annie’s DNA.” Leave room to glue the DNA strands from the other crew members below.
  8. Do the same for each of the other crew members’ DNA strands.
  9. Analyze your evidence.
  10. When you have glued down all the strands and labeled them, circle which one you believe is the unsanitary crew member that left his/her hair in the Egg McMuffin!
  11. Background: In 3-4 sentences, summarizehow youprepared your DNA evidence for analysis. Do not just copy these steps.
  12. Conclusion: Write 1-2 sentences describing how you knew which crew member was the culprit. REMEMBER, you cannot actually read the DNA sequence!
