6. BRIEF RESUME OF THE INTENDED WORK:
6.1 Need for study:
Among the eight branches of Ayurveda, Rasayana Tantra holds an important place to mention specially. Rasayana Tantra as the name suggests describes the methods of withdrawing ageing, increasing lifespan, intelligence, strength and capacity to get rid of diseases1. To be specific Rasayana is that by which one attains the excellence of all Dhatus2.
Vayasthapana is one among the qualities of Rasayana. Vayasthapana means maintaining the Tarunya Avastha1. Acharya Charaka has mentioned Vayasthapaka Gana3. Classics have also mentioned Ghrita, Ksheera, Sheetodaka and Madhu serve the similar purpose of Vayahsthapana. The effect of Vayasthapana can be achieved by taking all these drugs indigenously, or in combinations4.
Ghrita is mentioned as Medhya and Rasayana5. Ksheera is wholesome for all living beings and Rasayana by nature6. Acharyas explained that milk and ghee, if taken daily gets the best out of rejuvenation therapy7. These two are being used by people as a food constituent and also as an effective medicine from the time unknown. To live a long, happy and disease free life is the basic desire of every human being. Thus the study is taken to assess the Rasayana property with special reference to Vayasthapana effect of Ghrita and Ksheera.
6.2 Review of the literature:
A person undergoing Rasayana therapy attains memory, intelligence of lustre, complexion and voice. The drugs, diet, regimen which promote longevity by preventing ageing and diseases are called Rasayana8.
Vayasthapana is what we desire to attain by Rasayana karma thus making it one of its Guna. Ghrita, Ksheera, Sheetodaka and Madhu are four age- sustaining remedies if taken singly, in combination of 2 or 3 or all of them4. Ghrita is the best Snehadravya and Vayasthapaka9. Ksheera is also having Vayasthapana and Rasayana Guna6. Milk and ghee, if taken daily gets the best out of rejuvenation therapy7. It is also mentioned that Ksheera and Ghrita is more beneficial against ageing when used together10.

6.3. Aim of the study:
To study the Rasayana w.s.r. to Vayasthapana effect of Ghrita and Ksheera.
6.4. Objectives of the study:
1. To study critically on Rasayana w.s.r. to Vayasthapana effect of Ghrita and Ksheera.
2. To evaluate the Vayasthapana efficacy of Ghritha and Ksheera through animal experimental study.
7. MATERIALS AND METHODS
7.1 Source of data:
Literary source:
· Review of all available classical texts on Rasayana w.s.r. to Vayasthapana effect of Ghrita and Ksheera.
· Articles from journals and other published works.
· Related source of data from the website.
Experimental source:
Wistar strain rats taken from Experimental Lab of S.D.M. College of Ayurveda, Udupi.
7.2 Method of collection of data:
Wistar strain rats of middle (9-10 months) and old (>12 months) age will be selected from Experimental Lab of S.D.M. College of Ayurveda, Udupi. Selected middle aged rats shall be randomly placed under 2 groups and old aged rats shall be randomly placed under 2 groups, so in each group minimum of 6 rats will be included.
EXPERIMENTAL METHODOLOGY:
· Middle aged rats: 2 groups will be made.
One group will be given Ksheera and Ghrita.
Second group will be a control group will be given 0.5% gum acacia.
· Old aged rats: 2 groups will be made.
One group will be given Ksheera and Ghrita.
Second group will be a control group will be given 0.5% gum acacia.
Inclusion Criteria:
1. Healthy Wistar strain rats of either sex will be considered.
2. Rats of middle age (9-10 months) and old age (>12months).
Exclusion Criteria:
1. Wistar strain rats weighing less than 120g.
2. Pregnant and diseased rats.
3. Rats used for and under trial of other experiments.
STUDY DESIGN
An experimental study wherein minimum of 12 rats in each of middle and old age will be selected.
DOSE
Route of administration- Oral.
Rat dose will be calculated on the basis of human dose by using standard conversion method.
Human dose × 0.018 × 5 = g /kg body weight in rats.
Duration of study:
Duration of administration - 30 days

2. Physical Strength –
1) Rotarod performance.
2) Grip strength.
3. Mental Strength –
Behavioral despair
4. Memory Experiments -
Moris water maze experiment.
5. Biomarkers –
1) D.H.E.A ( Dehydro epiandrosterone)
2) Status of oxidative and antioxidative systems in brain.
- lipid peroxidation
- catalase activity
- glutathione content of brain
6. Body weight measurements.
7.3 Does the study require any investigation or intervention to be
conducted on patient or other humans or animals? If so please
describe briefly.
Yes, the study will be conducted on animal models.
