Supp. Fig. 1 The growth pattern in unirradiated cells measured as cell index by xCELLigence RT-CIS in different serum concentrations produces a similar growth pattern for each cell line. xCELLigence RT-CIS growth curves in unirradiated cells a. EMT6.5 b. NMUMG c. 4T1.2 and d. WTMacs propagated in DMEM media supplemented with 2% (red), 5% (green) and 10% Blue) serum (Data: Mean ± S.E.M, n=4). Each cell line produced a specific pattern of the growth curve. Media was chosen to be supplemented with 2% serum since it produces a low steady state of growth with 2 - 4 days of monitoring.** There are some error bars that are smaller than the symbol .
Supp. Fig. 2 Low plating efficiency was observed in 2% and 5% serum compared to 10 % serum. Colony number formation in unirradiated cells from clonogenic assay a. EMT6.5, b. NMUMG c. 4T1.2 and d. WTMacs, seeded at different cell seeding densities per plate and grown in media containing 2%, 5% and 10% serum (Data: Mean ± S.E.M, n=3).
Supp. Fig. 3 Cell proliferation measured by cell index using xCELLigence RT-CIS and absorbance by classic CTB assays produced a similar growth pattern. Dynamic proliferation of EMT6.5, 4T1.2 and NMUMG measured as cell index using xCELLigence RT-CIS (Left panel) and as absorbance using classic CTB assays (right panel) at similar seeding densities in unirradiated cell for six days (Data: Mean ± S.E.M, n=3). Cells were seeded with same number for each well for both assays and the proliferation patterns was compared.** There are some error bars that smaller are than the symbol
Supp. Fig. 4 EMT6.5 cell morphology showing enlarged cells after 7.5 Gy 3 days post irradiation. Same number of control and irradiated (7.5 Gy) cells were seeded in 6 wells plate and the morphology was observed for 4 days. Scale bar= 50 µm.
