Procedure:
- Get four bottles filled with algae N. oculata that have been in the dark for several minutes (Mr. Koons will take care of this).
- Measure the (initial) amount of dissolved oxygen using the dissolved oxygen (DO) sensor attached to the LabQuest. Carefully follow these directions.
b)Carefully raise the DO sensor out of the distilled water (be careful not to damage the glass end of the sensor) beaker and set aside.
c)Carefully lower the sensor into the liquid in your algae bottle.
d)Gently swirl the bottle - it’s important that the water is moving around the sensor to get an accurate reading.
e)Wait until the reading “stabilizes” (do not wait more than 30 seconds and write the number shown in your data table (initial)).
f)Write down what units are being used (mg/L)
- (Data collection is not a mindless activity. Notice if any of the data looks unexpected (the readings showed be between 4 and 10)
- Cap each bottle and place one under each of the different light conditions for 15 minutes.
- Measure the dissolved oxygen after 15 minutes (final), using the direction in step 2 –use the same DO sensor that you used before.
- Calculate the change in dissolved oxygen. This is a measure of the dissolved oxygen produced by photosynthesis in algae under strong light in 15 minutes.
- Enter your data into the spreadsheet on the Koons’ computer to get class data.
