Supplement 2. Flow cytometric analysis of IL-17+ expression in activated T cells after mixed lymphocyte reactions used in this study.

After DE induction, 16 corneas from DE-induced group were secured, and CCR7+CD11b+ and CCR7-CD11b+ cells were isolated by cell sorting FACS analysis. IFN-γ-IL-17-CD4+ T cells were harvested from submandibular and cervical LNs of normal condition group and sorted by FACS analysis. To measure T cell activation, each CCR7+CD11b+ and CCR7-CD11b+ cells were thoroughly washed and subsequently co-cultured with IFN-γ-IL-17-CD4+ T cells. After 5 days of incubation, samples were stimulated with PMA/ionomycin for 10 h with GolgiPlug. Responder T cells were collected for subsequent flow cytometry analyses of intracellular IL-17 expression. After permeabilization, intracellular cytokine levels were measured with anti- mouse IL-17-FITC antibodies.
