INTENDED USE

One of the aims of diary cow feeding management is to improve milk output and shortening the period of no pregnancy.

PRINCIPLE

Detection of pregnancy in dairy cow offers valuable information to producers. For a long time, dairy cow feeders and veterinarians are looking forward to simple, rapid and effective early pregnancy detection method, which is important for improving feeding management and propagation rate. If the pregnancy is not promptly detected after mating, it will result in delaying the period of calving and decrease of propagation rate and milk output. Thus the cost of feeding management will be increased. If the pregnant dairy cow can be detected in early period, the cow can get careful nursing and gestation management, which will increase the pregnancy success rate. Non-pregnant cow can be hybridized early, shortening the period of non-pregnancy and decreasing the feeding cost. Thus propagation rate and economical benefit will be increased. Study on the diagnostic skill for early pregnancy of diary cow is very important to the development of the dairy cow industry.

Progesterone (P4) is a kind of steroid hormone. The concentration change of P4 in milk is one specific characteristic indicating the activities of cow ovary. The estrous cycle of dairy cow is 21 days on average. P4 is in low-level at the beginning of the estrous cycle, thereafter rise gradually, reaches more high-level in middle period of estrous cycle, P4 level in non-pregnant cow begins to drop to the lowest level on the 17th day after mating, while it is still at high level in pregnant dairy cow's blood and milk. This product is designed and developed on the basis of the estrous cycle law of dairy cow. It can be performed at the end of estrous cycle. If only one red line appears, it indicates that the cow is not pregnant; P4 is in the lowest level at this moment. It is recommended to make the best use of this time to have the diary cow mate; If two red lines appear, it indicates the cow is pregnant. It should get careful nursing and gestation management.

The test theory is based on variation of P4 in the estrous cycle, to determine whether the early pregnancy of dairy cow by detecting P4 in the end of estrous cycle .The test strip employs colloidal gold immunochromatographic technology for early pregnancy diagnosis of diary cow, with characteristics of high accuracy, easy to operate, fast result, room temperature storage, easy to carry, safe to mother and baby cow.

SCOPE OF USE

The test can be used to determine the early pregnancy of diary cow, reexamine diary cows, and guide the artificial fertilization at right time.

Specificity: 97.8%

Analytical Sensitivity: 2ng/ml

Sensitivity: 96.9%

Specificity of this test was determined from cross-reaction studies with FSH (100μg/mL), E2 (10μg/mL), PRL (100μg/mL), LH (25mIU/ml), DHT (10μg/mL), tetraborane (10μg/mL) and H2O. No cross reactivity was observed.

HOW TO USE

Because the diary cow’s estrous cycle is 18-22days, it is suggested to perform the test on day 21st and again on day 24th after breeding:

1. If the test results from day 21st and day 24th are positive the cow is pregnant. NO NEED TO TEST AGAIN.

2. If the test results from day 21st and day 24th are both negative, the cow is not pregnant. NO NEED TO TEST AGAIN.

3. If the test results from day 21st is negative, you must still perform the test on day 24th to make sure the cow is not pregnant.

4. If the test results from day 21st are negative and the test results from day 24th are positive, perform the test again the following day (preferably with urine) to determine if the cow is pregnant or not.

5. If the test results from day 21st are positive and the test results from day 24th are negative perform the test again the following day (preferably with urine) to determine if the cow is pregnant or not.

6. Beyond day 24th, perform the test between days 40th and 45th for 5 continuous days.

SPECIMEN COLLECTION AND PREPARATION

Take whole milk to test its progesterone level directly. In order to get reliable result, give up the first three milking and then collect sample milk around 5-10ml. Mix the sample milk well before testing.

Note 2: The test works with eithera sample of milk, blood, or urine.Use themost practicalspecimenfor your operations incaseof heifers(i.e.,urine or blood.) The instructionsapply equallyto all three typesofspecimens.

Note 3 (Blood specimen): Collect the blood sample in a collection tube and leave the tube in horizontal position for 30 minutes, then in a vertical position for another 30 minutes, until blood clot. A clear liquid appear over the clot. Collect the serum in the dropper and pour 3-4 drops on the circular well.

ASSAY PROCEDURE

1. Remove the test device and milk dropper from its foil wrapper by tearing along the slice.

2. Using the milk dropper, withdraw the milk sample from the specimen cup and slowly dispense 3 – 4 drops into the circular sample well. The sample liquid moves slowly to the other end due to the capillarity. You will see one or two red lines in the result window.

3. Read results at 5 minutes.

INTERPRETATION OF RESULTS

Positive: If both T band and C band are visible, the test result is Positive, which means the cow is pregnant or in middle period of estrous cycle.

Negative: If only C line is visible, the test result is negative, which indicates the cow is not pregnant.

Invalid: When control line does not appear on the membrane, the test is invalid due to improper test procedure or deterioration of reagents. It is recommended that the test be repeated.

PRECAUTION

(1) Test according to the law of the estrous cycle strictly.

(2) For single use only; do not reuse the test devices.

(3) Don’t keep the strip or cassette in the air for a long time after unpacking.

(4) While performing the test, make sure that the surface of sample should not exceed the “MAX” line of strip.

(5) Do not use test kit after expiry date.

VALIDITY AND STORAGE

The period of the validity is 24 months. Store as packaged in the sealed pouch under the room temperature.

Avoid light.
