Diagnostic reagents and controls for perfoming Equine rhinopneumonitis serological assays (ELISA, CFT, HI, VN) and moleculal tests (nested PCR) were produced and tested in-house at a quantity sufficient to test all samples in the reporting period.

Our laboratory has supplied reagents for ELISA and HI to veterinary diagnostic laboratories within Russia and to 2 other OIE Member countries.

Part II: Activities specifically related to the mandate
of OIE Reference Laboratories

3. International harmonisation and standardisation of methods for diagnostic testing or the production and testing of vaccines

___

4. Preparation and supply of international reference standards for diagnostic tests or vaccines

___

5. Research and development of new procedures for diagnosis and control

This year the project on the development of recombinant construction pIYT.1 coding gene gD EHV-1 has been completed. Comparative analyses between pIYT.1, live and inactivated vaccines were performed using the laboratory model (BALB/c mice). The results showed that antibodies produced to the pIYT.1 construction could be identified in ELISA, NT and HI at the level similar to the antibody level induced by the live and inactivated vaccines. The live vaccine (EHV-1 attenuated strain “CB/69”) in optimum dose provides complete protection to challenge with neuropathogenic adapted in mice EHV-1 strain. The live vaccine (EHV-1 attenuated strain “Rhinomune”), inactivated vaccine and pIYT.1 provides incomplete protection to experimental challenge. Combined immunisation by the recombinant construction and several vaccines increase humoral immunity and protection to experimental challenge in mice. The pIYT.1 also induced antibodies to the EHV-1 in 1 – 1,5 year old foals.

A preclinical test with the developed inactivated vaccine against equine rhinopneumonitis and Salmonella abortus equi was performed. The laboratory results showed that the vaccine provided protection to equine rhinopneumonitis experimental challenge in 87,5% immunized mice. Preliminary tests of this vaccine were performed on pregnant mares of Yakut breed (endemic for Eastern Siberia region of Russia). 18 mares were vaccinated and 16 mares were used as a control. Healthy foals were born in vaccinated group (82,3%) and in control group (66,0%). This project has been performed in collaboration with Yakut Research Agricultural Institution.

The ORF30 identification of EHV-1 isolates showing a high risk of paralytic disease is in progress.

6. Collection, analysis and dissemination of epizootiological data relevant to international disease control

Collection, analysis and dissemination of epizootiological data relevant to international disease control has been performed in the connection of import of horses to Russia from several countries (Mongolia, Poland and countries of the former Soviet Union). Antibodies to Equine Rhinopneumonitis in NT were detected in 72 % samples.

The five – year scientific project on research of the circulation of viruses of domestic animals in marine mammals has been completed. This project was performed in collaboration with zoologists from Russian Agricultural Academy Timiryazev in Kurile Islands (Far East region of Russia). Antibodies to EHV-1 and Equine Influenza virus H7N7 has been detected in ELISA in more 50% cubs of Steller sea lions (n=44).

7. Provision of consultant expertise to OIE or to OIE Members

Experts from our laboratory (Prof. K. P. Yurov DVM and S. V. Alexeyenkova DBSc) provides advice on Equine rhinopneumoniis diagnostic test procedures and vaccine production to specialists from Russia, Ukraine, Kazakhstan on International Conference of Russian Equine Veterinary Assosiation in Moscow, International Conference “ Problems in horse breeding” in Novosibirsk, All – Russian Conference Directors of Regional Veterinary Laboratories in Kaliningrad.

Our laboratory carries out international mission to test horses on OIE equine viral diseases list in export/import and transit of horses through Russia.

8. Provision of scientific and technical training to personnel from other OIE Members

Technical training on laboratory diagnosis methods was given to Dr. M. M. Saidarova (Tajikistan).

9. Provision of diagnostic testing facilities to other OIE Members

Since 1977 until now our laboratory has provided official diagnostic services in export/import of horses through Russia to different countries (this year Mongolia, Chine, Iran, countries of EU and former Soviet Union).

10. Organisation of international scientific meetings on behalf of OIE or other international bodies

We were organizers of 3th International Conference “Problems in horse breeding”, 6 – 7th October, 2010, Novosibirsk city, Eastern Siberia region, Russia. Our co – organizers were Russian Institution of Experimental Veterinary Medicine in Siberia and Far East region. The numbers of participants were 163 persons. Proceedings of 3th International Conference “Problems in horse breeding” were published on 176 pages.
