Abstract

Carvacrol (2-mehtyl-5-(1-methylethyl)-phenol) is a predominant monoterpenic phenol which occurs in many essential oils of the family Labiatae including, Origanum, Satureja, Thymbra, Thymus, and Corydothymus species. In our earlier study we investigated the hepatoprotective activity of carvacrol, using D-galactosamine (D-GalN) induction as a model of hepatotoxicity at three different doses (20,40 and 80mg/kg body weight). D-GalN- hepatotoxic rats exhibited elevation in hepatic marker enzymes such as aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, gamma glutamyl transpeptidase, Oral administration of carvacrol for 21 days reversed these parameters towards normalcy. The biochemical observations were supported by histological examination of rat liver and kidney tissue. The 20 mg/kg body wt. dose of the compound produced a better effect than 40 or 80 mg doses. These results suggest that carvacrol could afford a significant hepatoprotective effect against D-GalN induced rats.

Since it is 20 mg dose i.e. the lower dose, that showed better activity we would now like try lower doses. Further, using optimum dose we have proposed to investigate the lipid profile, lipidperoxidative markers such as thiobarbituric acid reactive substances and lipid hydroperoxides and the activities of enzymatic antioxidant enzymes superoxide dismutase, catalase, glutathione peroxides, and nonenzymatic antioxidants (vitamin C, vitamin E and reduced glutathione) in the plasma, erythrocytes and tissues in carvacrol- treated GalN-induced hepatoxic rats. We will examine PPAR-alpha and gene expression by RT-PCR and the inflammatory markers such as COX-2 & TNF-α protein expression by western blotting analysis upon carvacrol treatment. We will also investigate the localization of cytokines such as IL-6 and cytochrome P450 by immunohistochemical analysis in the liver after carvacrol treatment. These results will be compared with silymarin the standard drug used for hepatotoxicity.
