Regulation of eukaryotic gene expression in the nervous system. Molecular basis of tolerance and physical dependence resulting from chronic opiate drug use. Molecular cloning of G protein-coupled receptors. Solubilization and purification of G protein-coupled receptors. Structure/function analysis of opioid receptors. Signal transduction pathways activated by opioids. Opioid receptor turnover, agonist-induced receptor down regulation, antagonist-induced receptor up-regulation. Ubiquitination and targets of the proteasome complex. Analysis of receptor post-translational modifications and interacting proteins by mass spectrometry. Select opioids ligands as pharmacological chaperones that stabilize and induce proper receptor folding during synthesis in the secretory pathway. Naltrindole and structural analogs as anti-cancer therapeutic agents.
