6 BRIEF RESUME OF THE INTENDED WORK
6.1 Need for the study
Science is not stagnant; it is always a continuous process. It needs scientific validation. Ayurveda is ancient Indian art and science of healing and rejuvenation. Rasashastra is the offshoot of Ayurveda. In Rasashastra different methods of unique preparations are mentioned among them khalveeya rasayana is one.
Khalvi method is a unique method applicable to most of the rasoushadhies. It brings specific changes in the minerals which accelerates the therapeutic action and help in better absorption.
Kamadugha Rasa is one such important formulation containing Mukta Bhasma, Pravala Bhasma,Mukta Shukti Bhasma, Varatika Bhasma, Shankha Bhasma, Shuddha Gairika, Guduchi Satva. It is indicated in Amlapitta, Pittaja vikara, Jirna Jwara, Bhrama1.It is also advised in Vruna due to Amlapitta2.
Peptic ulcer is a world wide problem. Peptic ulcers are the areas of degeneration and necrosis of gastrointestinal mucosa exposed to acid-peptic secretions. Though they can occur at any level of the alimentary tract that is exposed to hydrochloric acid and pepsin, they occur most commonly (98-99%) in either the duodenum or the stomach in the ratio of 4:13. Current statistical data shows more than 70% of people are suffering from gastro-intestinal disturbances.
For the purpose of globalization of Rasoushadhies, preclinical methods like experimental study are very essential.Hence keeping the above as asset this study entitled “PHARMACEUTICO - ANALYTICAL STUDY OF KAMADUGHA RASA AND ITS ANTI –ULCER ACTIVITY ON ALBINO RATS - AN EXPERIMENTAL STUDY” is undertaken.
6.2 REVIEW OF LITERATURE
- Preparation of Kamadugha Rasa will be reviewed as per different Ayurvedic texts.
- Review of Shodhana of Mukta4, Pravala5, Mukta Shukti6, Varatika7, Shankha8and Gairika9.
- Review of Marana of Mukta10, Pravala11, Mukta Shukti12, Varatika13and Shankha14.
- Review of Preparation of Kamadugha Rasa1.
- Review of concept of Amlapitta, Parinama shoola,annadrava shoola and their treatment as per Ayurvedic classics.
- Review of concept of Peptic ulcer as per modern science.

6.3 OBJECTIVES OF THE STUDY
- To carryout Shodhana of Mukta, Pravala, Mukta Shukti, Varatika, Shankha and Gairika.
- To carryout Marana of Mukta, Pravala, Mukta Shukti, Varatika and Shankha.
- To prepare Kamadugha Rasa.
- To do physico-chemical analysis of Kamadugha Rasa
- To evaluate anti ulcer activity of Kamadugha Rasa on albino rats by Indomethacin(20mg/kg) induced ulcer method.
7. MATERIALS AND METHODS
7.1 Source of data:
Literary:
All the relevant classical and up to date literature of Kamadugha Rasaand its
ingredients will be screened.Experimental anti ulceractivity study will be reviewed as per
modernPharmacology.
Pharmaceutical:
Genuine raw materials will be collected from authentic source.These will
be screened for Grahya lakshanas.Necessary processing of raw materials will be done in
Pharmacy section of P.G.Department of Rasashastra,in Taranath Govt.Ayurvedic
MedicalCollege, Bellary.
Analytical:
Kamadugha Rasa will be taken for physico-chemical analysis with
collaborationof accredited laboratory.
Experimental:
Albino rats will be usedfor experimental study.Ranitidine Hydrochloride
willbe used orally for the standard group.Animal experimental study will be carried out
at thedepartment of Pharmacology,Vijayanagar Institute of Medical Sciences (VIMS),
Bellary and the albino rats required for thestudy willbe obtained fromThe Central
AnimalHouse,VIMS,Bellary.
7.2 METHODS OF COLLECTION OF DATA
Pharmaceutical Study:
- The raw materials will be screened and data will be collected.
- Purification of Mukta will be done by subjecting it to swedana process in dolayantra with agastya swarasafor 1 yama4.
- Purification of Pravala will be done by subjecting it to swedana process in dolayantra with sarjika ksharajala for 1 yama5.
- Purification of Shukti6, Varatika7,Shankha8 will be done by subjecting it to swedana in dolayantra with nimbuswarasa.
- Purification of Gairika will be done by subjecting it to godugdha Bhavana9.
- Preparation of Mukta bhasma will be done by subjecting shodhita mukta to 3 laghuputa10.
- Preparation of Pravala bhasma will be done by subjecting shodhita pravala to 3 gajaputa11.
- Preparation of Shukti bhasma12, Varatika bhasma13, Shankha bhasma14 will be done by subjecting shodhita Shankha, Shukti, Varatika to puta.
- Kamadugha Rasa is prepared by triturating equal quantities of Mukta bhasma, Praval bhasma, Mukta Shukti bhasma, Varatiika bhasma, Shankha bhasma, Gairika and Guduchi Satva in the khalva yantra1.
Analytical Study

Experimental study:
Animal- Wister strainalbino rats
Drug - Kamadugha Rasa
Method- Indomethacin (20mg/kg) induced ulcer model15
Method:
The albino rats of either sex are randomly distributed into 3 groups of 6 animals each.
Group- I: Serves as control group which receives Gum acacia.
Group-II: Serves as standard group which receives Ranitidine Hydrochloride
Group-III:Serves as trial group which receives Kamadugha Rasa.
The control drug, standard drug and trial drug will be given once a day for 6days, orally
to the albino rats of respective group. On 6th day night all the animals will be fasted
overnight. Next day the control drug, standard drug and trialdrug will be given 2hour
prior to the administration of ulcerogen i.e. Indomethacin orally. After 3 hours, animals
will be sacrificed by ether anaesthesia.Then the abdomen will be opened. Stomach is
dissected and screened for gastric lesions and ulcers.
Gastric lesions will be studied under compound microscope or with the help of hand lens
(10x). Ulcer index is calculated16.
Statistical analysis : Will be done by unpaired ‘t’ test
Mean ± standard deviation and diagrammatic representation will be utilized.
7.3 Does the study require any investigations or interventions to be conducted on patients

The present study is carried out on albino rats.The albino rats of either sex with 150-200gm are taken.The animals utilized for experimental study will be sacrificed for the purpose of study. The experiments are conducted under the guidance and supervision of Guide and Co-guide.

The ethical clearance has been obtained from The Institutional Animal Ethics Committee
(IAEC), Vijayanagara Institute of Medical Sciences, Bellary. Xerox copy of Ethical
